The purpose was to determine extent that individuals near spray irrigation sites are exposed to microorganisms in wastewater aerosols. Report covers Phase II - a monitoring effort from May 1976 to April 1977 of a spray irrigation site utilizing unchlorinated secondarily-treated wastewater from biofiltration treatment process. Objectives included an in-depth pathogen screen of wastewater, establishing the relationship between pathogen levels and traditional indicator organisms, monitoring microorganisms in air within 600 meters of the spray source, and development/validation of a microbiological dispersion model for predicting pathogen aerosol concentrations. Effluent was monitored for microbiological, chemical, and physical characteristics; extensive microorganism and dye aerosol samples were collected (77 aerosol runs). Enteroviruses were detected in air, but at a very low concentration. Conclusions: There is considerable underestimation of pathogen aerosol levels when using traditional indicators to predict human exposures to enteroviruses. Microbiological dispersion model may be used with minimal monitoring to estimate exposure. There is little correlation between wastewater levels of traditional indicators and pathogens. Microbiological wastewater aerosols are generated by spray irrigation, do survive aerosolization and can be transported to nearby populations

The research programme over this 1 1/2 year period has followed very closely our original proposal, where we aimed to isolate very early T precursors from the human thymus, to determine if they expressed CD4 as in the murine thymus, and then to test if they could be targets for HIV infection. However, several of our findings were unexpected, although still very pertinent to the current views on the development of AIDS. This will lead to some shift of emphasis in the second half of the grant period. We have prepared and conjugated all the monoclonal antibody reagents required for this previously murine T-cell laboratory to work on the human thymus. We have secured a reliable supply of infant thymus tissue. We have carried out a thorough flow cytometric analysis of human thymocyte populations, which has led to a paper describing several new observations. We have developed procedures to deplete human thymus cell suspensions of the more developed T-lineage cells, in order to obtain access to the very few early T precursors

The purpose of the research program is to define immune responses of humans to Human Immunodeficiency Virus (HIV) infection and the cause of the Acquired Immune Deficiency Syndrome (AIDS). Immunodeficiency caused by HIV infections may be due to immune mediated lysis (by antibody and/or HIV specific cytotoxic T lymphocytes CTL) of infected macrophages and T4 lymphocytes. The definitions of these immune responses to HIV virus are needed for rational approaches for therapy and prevention. Since the project began in October, 1986, we have developed an integrated program of research projects to develop experimental systems required for defining the immune responses of humans to HIV.
