The archaeal box C/D complex requires an RNA component possessing Watson-Crick complementarity to the target RNA in addition to three proteins: L7Ae, Nop5p, and fibrillarin. Interestingly, fibrillarin, the protein responsible for catalyzing the direct methyltransfer from S-adenosylmethionine to the target RNA, has no affinity for the sRNA or sRNA/L7Ae complex. Instead, fibrillarin is brought to the site of methylation through tight association with Nop5p which in turn binds tightly to the sRNA/L7Ae complex. Although the intermediary role of Nop5p is clear, the details of the interaction of Nop5p with the sRNA/L7Ae complex have not been elucidated. We have used alanine scanning to clearly reveal the existence of two separable Nop5p domains with one being entirely responsible for binding to the sRNA/L7Ae complex (termed Nop-RBD for Nop-RNA binding domain) and the other for binding fibrillarin. The isolated Nop-RBD folds correctly as demonstrated by X-ray crystallography and binds to the sRNA/L7Ae complex with near wild type affinity
