The Ninth Annual Pezcoller Symposium entitled "The Biology of Tumors" was held in Rovereto, Italy, June 4-7, 1997. It focused on the genetic mechanisms underlying het­ erogeneity of tumor cell populations and tumor cell differentiation, on interactions be­ tween tumor cells and cells of host defenses, and the mechanisms of angiogenesis. With presentations at the cutting edge of progress and stimulating discussions, this symposium addressed issues related to phenomena concerned with cell regulation and cell interactions as determined by activated genes through the appropriate and timely media­ tion of gene products. Important methodologies that would allow scientists to measure dif­ ferentially genes and gene products and thus validate many of the mechanisms of control currently proposed were considered, as were the molecular basis of tumor recognition by the immune system, interactions between cells and molecular mechanisms of cell regula­ tion as they are affected by or implemented through these interactions. The molecular and cellular mechanisms of tumor vascularization were also discussed. It was recognized that angiogenesis provides a potential site of therapeutic intervention and this makes it even more important to understand the mechanisms underlying it. Suppression of BRCAl transcription may contribute to the development of a subset of breast cancers. Previously. we described a positive regulatory region (PRR) in the BRCAl promoter (J. Biol. Cheni. 274,8837). Deletion of the PRR resulted in a significant loss of BRCAl promoter activity. In continuing studies we observed that the minimal BRCAl promoter extends from -202 to -156 (therefore excludes the start site). The promoter (with the PRR) primarily Consists of two discrete functional domains- (1) A polypyrimidinelpolypurine (Py/Pu) tract and (2) A CREB- (cAMP response element kinding) like binding site. Mutations in either of the domains severely reduced transcriptional and binding activity. Additional results strongly suggest that estrogen exerts its indirect, positive effect on BRCAl transcription through the PRR

Next, we study the role of ribosomal S6 kinase 2 (RSK2) which is a downstream effector of the Ras-extracellular signal-regulated kinase (ERK)/ mitogen-activated protein kinase (MAPK) signaling cascade. We show that miR-29a/c and miR-140-5p target and downregulate RSK2 at the protein level. Suppression of RSK2 by these miRNAs in turn is able to increase TRAIL-induced cytotoxicity and inhibit NF¿B activation. These observations suggest that targeting of RSK2 and regulation of its signaling may have significant therapeutic potential
Characterization of TCL1-murine B-1a cell transcriptome dynamics reveals novel insights into chronic lymphocytic leukemia onset by Yuntao Dai( )

Chronic Lymphocytic Leukemia (CLL) is the most common adult leukemia in western countries and is characterized by an abundance of malignant neoplastic B cells. The normal counterpart to CLL B cells is B-1a cells. In order to comprehend the systematic alteration occurring in the early transformation stage of CLL, we performed high-throughput sequencing analysis via next generation sequencing (NGS) of mRNA from malignant B lymphocytes and normal B-1a cells. T-cell leukemia/lymphoma 1-transgenic mice (TCL1 mice) and their wild-type counterparts (WT mice) were used as cell donors. We detected an increasing number of differentially expressed genes with progressive aging of the paired groups. Among these candidates, we identified protein-coding genes and noncoding genes (microRNA precursors, long non-coding RNAs and pseudogenes), many of which are of potential therapeutic value. In particular, Neto2 (neuropilin NRP and tolloid TLL - like 2), a transmembrane protein that modulates glutamate signaling, was found to be the most upregulated in the B-1a cells of TCL1 mice. Its human homolog, NETO2, is positively associated with the expression level of ZAP-70, an important prognostic marker in human CLL. The differential expression of selected representative candidates was validated in mature mouse CLL cells as well as in human CLL patients. In addition, the correlation of these candidates with TCL1 was verified with ectopic expression in human cell lines. The profiling dataset underlies future investigation for potential therapeutic targets. Among them, NETO2 is the first identified to be of potential clinical value as both prognostic marker and therapeutic target
