The main objective of this project is to determine conditions of exposure of human lymphocytes in vitro to continuous wave (CW) or pulsed wave (PW) 2450 MHz microwaves, which: affect the course of spontaneous or phytohemagglutin in (PHA)-induced lymphoblastoid transformation, and may induce numerical and/or structural aberrations in chromosomes of cells undergoing division following transformation. In order to provide reliable quantitative data on exposure, a system will automated dosimetry was developed, and tested for biocompatibility with lymphocyte cultures in vitro for periods of up to 120 hrs. (5 days). A method for quantitation of lymphocyte transformation based on image analysis was developed and tested. Experiments on effects of exposure on spontaneous transformation were carried out. For convenience and clarity the work performed is described under two separate headings: (1) exposure system and dosimetry, and (2) studies on lymphocyte cultures

This contract provided support for the procurement of equipment to build and operate improved near-field scanning optical microscopes (NSOMs). Improved scanning stages were constructed, and a scheme for high speed scanning developed based on distance regulation with tuning forks. In this scheme the phase of the drive to the fork is monitored with a digital phase-locked loop, which allows very rapid adjustment of the fork position in response to damping changes that occur as the fork and optical fiber tip are scanned in close proximity to a surface. In addition, new excitation and detection schemes were developed, which have allowed us to pioneer the use of nonlinear NSOM for the study of ferroelectric and ferromagnetic domains, and the crystal structure of surfaces

This report describes the development of both incoherent and coherent fiber optic sensors for monitoring the electrical activity of cells and tissue. The incoherent sensors use an optical fiber to deliver laser light to a cell membrane that is stained with a voltage-sensitive dye. The change in resulting fluorescence allows the electrical activity of the cell to be monitored. The coherent fiber sensor uses an external element, either a cell or some other material that has induced birefringence, as an extrinsic sensing element. The fiber is a means for delivering coherent light to this element. Birefringence- induced phase changes are detected in a heterodyne interferometric scheme. This report also describes the development work to date on NanoSQUID, a very small scale, superconducting quantum interference device that will allow spatially- resolved measurements of the magnetic activity of cells and tissue.
