Nuclei are dynamic organelles that are organized into several substructures. The perinucleolar compartment (PNC) is a substructure found in transformed cells that is apparently involved in RNA metabolism. Our studies of unknown antigens found in PNCs and in nucleoli characterized the localization and interactions of these antigens and indicated that these antigens may be important for maintaining nucleolar and PNC structure. Nucleoli are the substructures in which many of the steps of ribosome synthesis take place. Ribosome synthesis begins with the transcription of preribosomal RNA (pre-rRNA) and requires the essential transcription factor UBF. Our studies utilizing the knockdown of UBF in Xenopus laevis embryos indicate that UBF is involved in processes essential for embryonic development that are most likely different than its role in the transcription of pre-rRNA. Once pre-rRNA is transcribed, U3 small ribonucleoproteins (snoRNPs) are involved in the processing it and assembling new preribosomes. Our studies of U3 snoRNP proteins indicate that they are nucleocytoplasmic shuttling proteins and provide support for a new model of U3 snoRNP formation. Our studies of the U3 snoRNP protein hSof1 provide evidence that this protein is involved in processes that differ from other U3 snoRNP components. Together, these studies have provided new insights into the function, interactions, and dynamics of PNC-associated and nucleolar factors

During transformation, ribosome synthesis increases concurrently with the size and number of nucleoli. U3 snoRNA-associated complexes are involved in the cleavage of pre-rRNA and 4OS preribosome assembly and contain a number of proteins whose roles in the complex are unclear. Our research focused on characterizing the functional roles of human U3 snoRNA-associated factors by examining their localization and dynamics in live cells. We found that these factors can be divided into subclasses based on their localization and dynamics and showed for the first time that U3 snoRNA-associated proteins shuttle between nucleoli and the nucleoplasm and between nuclei and the cytoplasm. These studies have led us to propose a model for the assembly and function of U3 snoRNA-associated complexes. We also carried out more extensive studies of the U3 snoRNA-associated protein Sof1 which defined the protein domains necessary and sufficient for its localization, characterized its interactions, and disrupted its function. These studies have indicated that Sof1 has characteristics that distinguish it from other U3 snoRNA-associated proteins. Overall, these experiments have clarified the roles of Sof1 and other U3 snoRNA-associated factors in ribosome synthesis and have contributed to our understanding of ribosome synthesis as a whole

Depletion of yeast Sof1 inhibits pre-rRNA processing, consistent with a functional role in the U3 snoRNP. Our research focuses on characterizing the role of human Sof1 (hSof1) in ribosome biogenesis by examining its localization and dynamics in live cells and its interactions. hSof1, like fibrillarin, localizes to both the nucleolus and nucleoplasm. However, unlike fibrillarin, hSof1 is also in the granular component of nucleoli and responds differently to the inhibition of the transcription of pre- rRNA. In addition, hSof1 -GFP also exhibits a higher nuclear mobility than fibrillarin-GFP and is a nucleocytoplasmic shuttling protein
