The Biomarker Laboratory of the National Wildlife Research Centre has been providing cholinesterase activity measurements in wildlife tissues in support of the National Wildlife Toxicology Program since the early 1990s. This report contains detailed descriptions of the analytical methodologies used, and written in a format consistent with the CAN/CSA-Z-753-94 standard (Guidelines for the Documentation of Test Methods). Three methods are included, based on well established spectrophotometric techniques. The first is used for the analysis of plasma samples and brain homogenates with acetylthiocholine iodide as substrate, while the second is used with whole blood samples using propionylthiocholine iodide. The third method is used in conjunction with the first for the differential diagnosis of organophosphorus and carbamate insecticides poisoning

Induction of cytochrone P4501A is a sensitive biomarker of exposure to environmental contaminants such as polycyclic aromatic hydrocarbons. This report contains a detailed description of a fluorescent spectrophotometric method to measure the product of the reaction of the cytochrome P450 associated enzyme (7-ethoxyresorufin-O-deethylase or EROD) and total protein simultaneously, using a plate reader. Appendices contain the details for preparation of microsomes (small membrane-enclosed vesicles derived from endoplasmic reticulum of fractionated cells) by gel filtration, for a procedure adapted for smaller liver samples, and for preparation of microsomes by ultracentrifugation
