Mycophenolsäure (MPA), ein weit verbreitetes Immunsuppressivum, übt seine pharmokologische Wirkung durch unkompetitive Hemmung der Inosinmonophosphatdehydrogenase Typ II (IMPDH-II), ein Schlüsselenzym in der de novo Synthese von Purinnukleotiden aus. In vivo wird Mycophenolsäure zu einem pharmakologisch inaktiven Phenolglucuronid (MPAG) und einem pharmakologisch aktiven Acylglucuronid (AcMPAG) metabolisiert. In dieser Arbeit konnten enzymkinetische Untersuchungen über die photometrische Bestimmung der bei der Aktivität von IMPDH-II entstehenden NADH zeigen, daß AcMPAG wie MPA ein unkompetitiver Inhibitor der IMPDH-II ist, wobei das hemmende Potential ungefähr um den Faktor 6-10 über dem der MPA liegt und daß dabei durch Bindung an Albumin (HSA) schon im physiologischen Konzentrationsbereich eine Reduktion des hemmenden Potentials des AcMPAG erreicht wird. Mycophenolic acid (MPA) is a potent inhibitor of inosine monophosphate dehydrogenase (IMPDH), a key regulator of purine biosynthesis. MPA is frequently used as an immunosuppressant drug to prevent acute graft rejection for kidney, liver and lung transplantation. The use of MPA is associated with GI toxicity which is a problem to the patients, and a challenge for clinicians. The present study was undertaken to identify novel molecular targets of MPA using a proteomics approach. Two dimensional gel electrophoresis (2-DE) and mass spectrometry were used to identify proteome alterations in human embryonic cells (HEK-293) following exposure to therapeutic concentrations of MPA. Cells were treated for 72 hours, and total cell lysate was resolved by 2-DE followed by QTOF-MS/MS analysis of all identified differentially regulated proteins. A total of 12 proteins were differentially regulated in HEK-293 cells following exposure to MPA. Im Rahmen dieser Arbeit wurde das antifibrotische Potential von Mycophenolat-Mofetil (MMF) bzw. seines aktiven Metaboliten Mycophenolsäure (MPA) untersucht. Hierfür wurden einerseits ein in-vitro-Modell für renale Fibroblasten (COS-7-Zellen) und andererseits COL4A3-defiziente Mäuse als Tiermodell für eine progrediente renale Fibrose verwendet. Die in-vitro-Experimente bestätigten die inhibitorische Wirkung von MPA sowohl auf die Proliferation von Fibroblasten als auch auf ihre Funktion hinsichtlich ihrer Fähigkeit zum Wundverschluss und zur Kollagen-Kontraktion. Sowohl die Proliferationshemmung als auch die Funktionshemmung im Wundverschluss-Assay waren durch Zugabe von Guanosin/8-Aminoguanosin (G/8-AG) reversibel. In einem Kollagenmatrix-Kontraktions-Assay konnte der MPA-vermittelte Effekt jedoch nicht durch G/8-AG antagonisiert werden. Da die Wirkung von MPA bisher allein auf die Hemmung der de-novo-Synthese der Purinbasen durch Blockierung des Schlüsselenzyms Inosinmonophosphat-Dehydrogenase (IMPDH) zurückgeführt wird, zeigen diese Ergebnisse erstmals neben der IMPDH-abhängigen Inhibition der Fibroblastenproliferation und des Wundverschlusses eine bisher nicht beschriebene IMPDH-unabhängige Inhibition der Kollagenmatrix-Kontraktion
