Neurodegenerative diseases are progressive pathologies that affect the nervous system, causing the death of nerve cells. The best known and most frequent are Alzheimer's and Parkinson's disease, but there are others. All these diseases are characterized by the progressive loss of neurons of the nervous system, leading to cognitive, motor or perceptual complications. Parkinson's disease (PD) is caused by the degeneration of dopaminergic neurons of the substantia nigra and their nerve endings that normally release dopamine into the striatum. The two main risk factors common to neurodegenerative diseases are age and oxidative stress. Oxidative stress plays a central role in the pathophysiology of PD, but the mechanisms involved in controlling this stress in dopaminergic cells are not fully elucidated. Many studies show that selenoproteins play a central role in the control of redox homeostasis and cell protection, but the precise contribution of members of this family of proteins during neurodegenerative diseases is still unknown. Previous studies performed in our laboratory have uncovered the essential role of a new selenoprotein, selenoprotein T (SelT) in the processes of neuronal differentiation, but the role of this selenoprotein in neuroprotection was not known. We first showed that SelT, whose gene knock-out is lethal during embryogenesis, exerts a potent thioredoxin-like oxidoreductase activity. In a cellular model of dopaminergic neurons, represented by SH-SY5Y neuroblastoma cells, modification of SelT expression affects the level of oxidative stress and cell survival. Treatment of wild-type mice by neurotoxins targeting dopaminergic neurons such as 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) or rotenone induced massive expression of SelT in the nigro-striatal system, suggesting that SelT could protect these neurons under conditions of degeneration. On the other hand, this same treatment given in mice invalidated for SelT in the brain caused a parkinsonian syndrome with the appearance of motor symptoms, thus confirming that the presence of SelT must participate in the protection of dopaminergic neurons under conditions mimiking PD. The observed motor symptoms are associated with oxidative stress and marked degeneration of dopaminergic neurons. Similarly, we observed a decrease in the active form of tyrosine hydroxylase, resulting in reduced dopamine levels in the striatum of invalidated and neurotoxin-treated mice. These data show that SelT is essential for the survival and functionality of dopaminergic neurons in vitro and in vivo under the conditions of neurodegeneration mimicking PD. Finally, in patients with PD, we observed a considerable increase in SelT levels in the caudate-putamen but not in other cerebral structures. Together, these results uncovered the activity of a novel thioredoxin-like enzyme that protects dopaminergic neurons against oxidative stress and prevents the early onset of severe motor symptoms in animal models of PD. Our data indicate that selenoproteins such as SelT, whose levels are increased in PD play a crucial role in protecting dopaminergic neurons against oxidative stress and cell death, thus paving the way for the development of new neuroprotection strategies targeting these proteins in PD

Terpenes represent more than 80% of natural oils of Morocco, are of moderate cost and very plentiful in nature. These compounds are prone to be readily functionalized, and as their backbone includes one or more chiral centers, they are therefore potential precursors for the synthesis of chiral ligands without separation of the enantiomers. In this work, we have been interested in the synthesis of novel chiral ligands from natural terpenes. In particular, we have prepared amino alcohols, diimino and diaminodiols from [alpha]-pinene, and [alpha]-aminooximes, their ethers and their corresponding diamines from limonene. These new ligands were applied in asymmetric catalysis of hydrogen transfer of aromatic ketones with complexes based on transition metals, particularly ruthenium, in the presence of isopropanol as hydrogen source. These ligands in comparison with the literature prove to be efficient in terms of activity and are shown to induce medium to good enantioselectivities (enantiomeric excesses up to 80% have been obtained on some substrates). Further study on the reaction mechanism have been carried out specifically with the [alpha]-aminooximes ligands, as this work is presenting the first application of this new series of ligands in asymmetric catalysis

Modélisation et étude expérimentale du comportement thermo hydraulique des fluides frigoporteurs diphasiques by Mohamed Najib El Boujaddaini( )

This work concerns the experimental investigation and modelling of the thermo hydraulic behaviour of a new Iwo-phase secondary refrigerant, the paraffin slurry, flowing through a rectangular channel of a heat plate exchanger type. The paraffin slurries are made of millimetric bullets of paraffin, stabilized in an organic porous polymeric matrix, in suspension in water serving as a carrying fluid. The paraffin particles used contain 75% of paraffin called NORPAR®15 and 25% of a tri-block polymer of styrene with High Molecular Weight (HMW). The experimental results generated by the heat balances on the test sections of the experimental setup built in the Thermal Center of Lyon (CETHIL), highlight an important increase of the heat transfer coefficient, due to the particles presence in the carrying fluid. For a laminar flow of the paraffin slurry in the cold channel, an average multiplication by 1 .25 to 1.5 of the global heat transfer coefficient compared to the single-phase fluid was recorded for particles mass fractions of 6 to 12%. By regression of the experimental results, correlations for the local and average Nusselt number calculation for the laminar flows are proposed. The particularity of the presented correlations is their validity in the case of a pure fluid as we\l as for a two-phase fluid containing so\id particles. A model for the hydraulic and thermal behaviours studies of a Iwo-phase secondary refrigerant fluid during its cooling in laminar flow through a rectangular channel was developed it is based on the mixture model and to king the slip velocity into account. The evolution of the experimental and theoretical values for the fluid average temperature, the heat flow which crosses the walls and the heat transfer coefficient between the wall and fluid shows good agreement and the model is satisfactory since the variations never exceed 14%

Une technologie alternative dans le domaine du transport du froid est actuellement envisagée avec les fluides frigoporteurs diphasiques (liquide - solide). Celle-ci permet de réduire la charge du fluide frigorigène dans l'installation, en la consacrant à la production de froid, mais aussi de transporter en grande quantité de l'énergie frigorifique sous forme latente. Les coulis de glace sont des fluides frigoporteurs diphasiques constitués de particules de glace dispersées dans de l'eau ou des solutions aqueuses. Une étude énergétique globale d'une installation de froid dans son intégralité, permet d'évaluer les performances de ce type d'installation. Nous avons commencé notre travail par une étude du réseau de distribution de froid. Nous avons développé un modèle afin d'analyser l'intérêt énergétique de l'utilisation d'un fluide frigoporteur monophasique ou diphasique en prenant en compte la consommation énergétique liée à son écoulement. Le modèle prend en compte les pertes de charges singulières et régulières d'une boucle de fluide et permet d'en déduire la puissance de pompage correspondante. Dans la mesure où nous disposons d'un banc d'essais et qui a fait l'objet de compagnes de mesures, une analyse énergétique de toute l'installation a été faite, en considérant chaque poste de consommation et a permis la validation expérimentale du modèle de perte de charge. Par ailleurs, se pose la question quant au transfert de chaleur au sein des échangeurs situés sur de tels réseaux de froid et à leur comportement face au caractère instationnaire de la fusion de la glace. Un modèle numérique a été réalisé, donnant une bonne correspondance avec les résultats issus de l'expérience
