Les tauopathies, y compris la maladie d'Alzheimer, sont caractérisées par des dépôts de protéine tau anormale, appelés dégénérescences neurofibrillaires (DNF). Bien qu'il y ait une corrélation entre la présence de DNF, la perte neuronale et le déclin cognitif, le rôle exact des filaments de tau reste controversé. Pour comprendre le lien entre DNF et mort neuronale, nous avons développé une méthode d'observation des DNF dans le cerveau vivant de souris à travers une fenêtre crânienne. Nous avons employé la souche rTg4510 qui porte le transgene humain tau P301L, et caractérisée par le développement de DNF et de perte neuronale. Nous proposons un nouveau modèle dans lequel la protéine tau phosphorylée soluble est la forme toxique, l'activation de caspase accélère l'agrégation de tau, tau normale est également recrutée dans les agrégats, et la formation des DNF ne conduit pas à la mort neuronale immediate. Le mécanisme de propagation de la pathologie de tau dans le cerveau est toujours inconnu. Chez les patients Alzheimer, la pathologie de tau apparaît d'abord dans le cortex d'entorhinal, avant de se propager à l'hippocampe puis au cortex entier. Nous avons visé à comprendre la diffusion de la pathologie de tau en développant un modèle de souris transgéniques dans lequel l'expression de tau humain P301L est limitée au cortex entorhinal. Notre modèle recréé la progression des dépôts de façon hiérarchique, du cortex d'entorhinal aux secteurs de la mémoire. La détermination du rapport entre DNF et mort neuronale, et l'identification des mécanismes de propagation de la maladie, engendrent de futures directions pour le développement des stratégies thérapeutiques appropriées
