Two genes, mpa (Mycobacterium proteasomal ATPase) and pafA (proteasome accessory factor A) are important for the bacterial proteasome degradation pathway, resistance to nitric oxide, and pathogenesis. Biochemical studies have begun to uncover what roles Mpa and PafA play in proteasome degradation, however no work had been done to identify the role of the proteasome degradation pathway in virulence. We hypothesized that the proteasome may transcriptionally control genes that are involved in virulence. Microarrays comparing WT with pafA and mpa mutant strains revealed that about 60 genes were differentially regulated. We focused on a previously uncharacterized copper inducible regulon that was down-regulated in pafA and mpa mutants, and found almost exclusively in pathogenic Mycobacteria. One of the regulated genes, Rv0190/MT0200, is a paralogue of a recently characterized copper metalloregulatory repressor, CsoR. As is expected for a repressor, disruption of Rv0190/MT0200 led to the constitutive up-regulation of the five copper responsive loci. Additionally Rv0190/MT0200 protein specifically interacted with a palindromic DNA motif found in all five promoters. We named Rv0190/MT0200 ricR for regulated in copper repressor. A ricR mutant was hyper-resistant to copper, suggesting that the genes in the RicR regulon are important for resisting copper toxicity. Although toxic at high levels, copper is essential to most life, and as an intracellular pathogen, Mtb must maintain copper homeostasis while in the macrophage. This presents an interesting possible connection between copper, the proteasome and pathogenesis
