Bacteria are colonizers of various environments and host organisms, and they are often subjected to drastic temperature variations. Dickeya dadantii is a Gram-negative pathogen infecting a wide range of plant species. Soft rot, the visible symptom, is mainly due to the production of pectate lyases (Pels) that can destroy the plant cell walls. Production of Pels is controlled by a complex regulation system and responds tovarious stimuli, such as presence of pectin, plant extracts, growth phase, temperature or iron concentration. Although many studies have been carried out, the mechanisms of control of Pels production by temperature have not yet been elucidated. In bacteria, thermoregulation acting at the level of transcription initiation occurs usually both via transcription factors and DNA topology. We show that PecT, a previously identified repressor, is involved in the thermoregulation of the pel gene expression. Using in vivo Chromatin ImmunoPrecipitation (ChIP) coupled to quantitative RT-PCR(qRT-PCR), we reveal that PecT binding to the pel gene promoters is modulated by temperature. By manipulating the DNA topology in vivo, we further show that DNA supercoiling state is involved in the thermoregulation of pel gene expression by PecT. In addition, we show that the development of the pathogenicity of the pecT mutant according to changes in temperature is different from that of the parental strain. This report presents a new example of how plant pathogenic bacteria use transcription factor and DNA topology to adjust synthesis of virulence factors in response to temperature variation

Magnetotactic bacteria (MTB) are characterized by synthesizing magnetosomes and swimming along geomagnetic field lines. Multicellular Magnetotactic Prokaryotes (MMPs) is the most evolved MTB. Two morphotypes of MMPs have been discovered: spherical (sMMPs) and ellipsoidal (eMMPs). In this thesis, I revealed a high diversity of eMMPs from the Mediterranean Sea. Four new eMMPs species have been detected, extending the total number of identified eMMPs in the Mediterranean Sea from one species of one genus to five species of two genera. I also performed a comprehensive analysis of eMMP architecture. The eMMPs was composed of one layer of cells that surround a central lumen. The constituent cells are arranged axisymmetrically along the long axis of eMMPs and radial symmetric along the short axis. The juxtaposed membranes connect cells into a multicellular entity. The eMMPs reproduce through periphery-core unilateral constriction of constituent cells and unidirectional binary fission of the ellipsoidal MMPs. I have also identified two Nitrospirae MTB collected, for the first time, from marine environment. They have similar morphology, motility but different colors. They synthesized multiple bundles of bullet-shaped magnetite magnetosomes. The two novel Nitrospirae MTB species expand the distribution of Nitrospirae MTB from freshwater to marine

Les entérobactéries accumulent lors d'un stress hyperosmotique, du glutamate puis du tréhalose. E. chrysanthemi se distingue par l'accumulation, en plus du glutamate, de glutamine et de glucosylglycérate. Les gènes impliqués dans la biosynthèse du glucosylglycérate ont été mis en évidence et le mutant correspondant est affecté lors d'un choc hyperthermique. De plus le glucosylglycérate protège in vitro la glutamate déshydrogénase d'E. coli contre l'inactivation thermique, lui suggérant ainsi un rôle thermoprotecteur. Lorsqu'ils sont ajoutés dans le milieu, les osmoprotecteurs se subtituent aux solutés endogènes. Dans ces conditions, l'ajout de bétaïne inhibe la croissance du mutant bspA d'E. chrysanthemi.. L'absence de BspA autorise l'ouverture d'autres canaux mécanosensibles moins spécifiques, responsables de cette perméabilité. La mutagenèse de mscL et des 4 homologues MscS a permis de les impliquer lors du développement en conditions hyperosmotiques et de chocs hypoosmotiques

L'osmoprotection de Sinorhizobium meliloti par la GB, nécessite la présence d'un gène sodA fonctionnel. Un mutant sodA confronté à un stress hyperosmotique en présence de GB présente une auxotrophie partielle à la méthionine, dont la biosynthèse est assurée par deux voies impliquant soit la méthionine synthase soit la bétaïne homocystéine méthyl transférase. Ces deux enzymes sont inactivées suite à un stress salin ou oxydatif. L'absence du régulateur Hfq a un impact drastique sur l'établissement de la symbiose. La capacité d'un mutant hfq à coloniser le cordon d'infection est réduite et seules quelques bactéries parviennent à atteindre le primordium de nodule. A l'état de bactérie libre, en phase stationnaire de croissance, l'absence de Hfq se traduit par une sensibilité accrue au stress oxydatif. Le mutant Hfq est caractérisé également par une diminution de la synthèse de nombreuses protéines dont, ChvE, AgpA, GroES1. Ce travail souligne la nature pléiotropique du régulateur Hfq chez S. meliloti aussi bien sous forme libre que symbiotique
