PSD-95-like membrane associated guanylate kinsases (PSD-MAGUKs) are multidomain scaffolding proteins involved in AMPA and NMDA receptor trafficking and clustering at synapses. As proteins with multiple protein-protein interaction domains, they have a capacity to interact with many integral membrane receptors, cytoskeletal components, and signaling molecules although how these interactions are regulated is unclear. Multiple conformational states for some PSD-MAGUKs have been described and could act to regulate the availability of their individual domains. However, an in vivo demonstration of these conformations, especially in neurons, and an understanding of what defines their conformational states is lacking. In the work presented here, conformations of PSD-MAGUKs and how they are regulated were inverstigated using a FRET-based approach where the N- and C-termini of SAP97beta, PSD-95alpha and SAP102 were tagged with a mCherry and Venus FRET pair. By measuring changes in intramolecular FRET, we demonstrate in HEK293 cells and in neurons that PSD-MAGUK proteins alter between "compact" and "extended" conformations. Further, we reveal that interactions between PSD-MAGUKs and different NMDAR and AMPAR subunits are dependent on PSD-MAGUK conformation. These studies also find that transitions between conformational states are regulated by changes at different sites at the PSD-MAGUK N-termini, the L27 protein interaction domain (SAP97beta), the palmitoylation site (PSD-95alpha), and the zinc-binding domain (SAP102). Parallel electron microscopy measurements at the PSD confirm and extend the conclusions about PSD-95alpha conformation based on FRET measurements. Our results reveal the dynamic nature of the structure of PSD-MAGUKs, identify a novel regulatory role for their N-termini, and provide mechanistic insight into how PSD-MAGUK family proteins determine their selection of different binding partners

At that time, executives rose almost exclusively within a single firm, learning the business over many loyal years of service. Since the 1970s, firms have progressively relied more on external markets for filling its leadership ranks. As a result, the value of executives has become increasingly defined by capabilities portable across organizational settings. External markets have less information about executive abilities compared to incumbent employers, which strengthens the influence of externally observable signals of quality on executive career opportunities and compensation. Across three studies, this dissertation empirically explores how external markets value executive human capital attributes. In particular, this work focuses on how external markets differ from incumbent employers and explores implications for executives building their careers across multiple organizations

The classification of consumable media by mining relevant text for their identifying features is a subjective process. Previous attempts to perform this type of feature mining have generally been limited in scope due having limited access to user data. Many of these studies used human domain knowledge to evaluate the accuracy of features extracted using these methods. In this thesis, we mine book review text to identify nontrivial features of a set of similar books. We make comparisons between books by looking for books that share characteristics, ultimately performing clustering on the books in our data set. We use the same mining process to identify a corresponding set of characteristics in users. Finally, we evaluate the quality of our methods by examining the correlation between our similarity metric, and user ratings

Increasing the speed of single-core processors has been facing practical challenges. Instead, multi- core architecture has been ascending for the past decade as the dominant architecture. To gain full advantage of multi-core processors, it is unavoidable for programmers to write concurrent programs. However, writing and reasoning about concurrent programs is often difficult for programmers. One reason for the difficulty stems from the hurdle of dealing with concurrency abstractions, the other reason is the difficulty in getting rid of concurrency related bugs. To address these problems, a new abstraction for concurrent programming has been proposed called capsule. Capsules are inspired by the long- standing ideas explored in the context of message-passing concurrency (MPC) abstractions and other similar models. Although the jury is still out on MPC abstractions as the de facto abstraction for concurrency, their wide availability and advantages combine to warrant research on the use of this model for concurrency. Capsules explore a new point in this design space to balance flexibility and analyzability in the MPC programming models. Unlike common avatars of the MPC model, a capsule is statically deployed and configured, confines its local states, permits only a single activity within itself, and communicates with other capsules in a logically synchronous manner. This thesis focuses on the realization, applicability and performance of this new abstraction. A major contribution of this work is the realization of capsules. We have implemented capsules as an extension of javac, the industrial strength standard Java compiler. The implementation shows that it is feasible to extend an object- oriented language with capsules. The default compilation strategy of capsules is based on threads. In this work, we also show alternative compilation strategies to improve the flexibility and adaptability of capsules. This shows that the capsule abstraction can be decoupled from concre
