This thesis is composed of two major areas of study: the first study describes yeast two hybrid screen and biochemical approaches which were performed to identify interaction partners for C. elegans CUL-3; the second study focuses on the functional analysis of a novel ubiquitin conjugating (UBC) domain-containing protein FTS (Fused Toes). In the first study, a large family of proteins harboring a conserved BTB domain was identified to partner with Cul3. The BTB domain adopts a structure similar to the Cul1 interacting domain of Skp1. Most of the identified BTB domain proteins also contain an additional potential protein-protein interaction domain, such as MATH domain. We hypothesized based on analogy with the SCF complex, that BTB proteins function as substrate specific adaptors of the Cul3 Ub ligase. Mel-26, an essential MATH-BTB protein was one of the BTB proteins identified from the screen that interacts with CUL-3 in both yeast two hybrid and 293T transfection systems. Previous studies indicated that MEI-1, a microtubule-severing protein, is negatively regulated by MEL-26. We found MEL-26 interacts with MEI-1 through its N-terminal MATH domain and interacts with CUL-3 via its C-terminal BTB domain. We proposed that the BTB domain family of proteins merge the function and properties of Skp1 and F-box proteins into a single polypeptide, thereby acting as substrate specific adaptors for Cul3

Abstract : We designed a series of 2D CMPs and found that the Fe-CMPs possess the highest catalytic activity toward CO2 photoreduction. Abstract : Herein, by means of density functional theory (DFT) calculations, we designed a new type of thiophene-linked porphyrin unit and incorporated it into an extended 2D conjugated microporous polymer (CMP) to serve as a CO2 reduction catalyst. This newly designed catalyst combines the advantages of molecular catalysis, heterogeneous catalysis, high structural stability and tunable electronic properties. A series of metal atoms (Fe, Mg, Mn, and Cu) were introduced into the center of the porphyrin ring to modify its capacity for reducing CO2 . Our transition state (TS) search calculations revealed that the Fe-modified CMP possessed the highest catalytic activity toward CO2 reduction. The plausible reduction pathway was also determined. The band structure and frontier orbital distribution calculations further demonstrated its semimetallic property and higher stimulated transition probability, both of which are critical factors for photocatalytic performance. We hope that our design could provide meaningful guidance for other relevant CO2 catalytic reactions

Isomeric Effects of Solution Processed Ladder-Type Non-Fullerene Electron Acceptors( )

Abstract : The role of electronic structure and thin film morphology is investigated in determining charge transfer and electron coupling due to orbital interactions in two isomeric non-fullerene acceptors with the structure of acceptor-donor-acceptor. Differences are found in the distribution of electron density of the highest occupied molecular and lowest unoccupied molecular orbitals, whose bonding interactions result in improved intermolecular interactions and hence, molecular stacking. When combined with a large band gap polymer donor, solution-processed organic photovoltaic cells are demonstrated with power conversion efficiencies as high as 10.5 ± 0.4%, and with absorption extending to wavelengths of 800 nm. Due to strong internal organization driven by the planar molecular structure and strong intermolecular interactions, no post-deposition processing such as solvent vapor or thermal annealing is required. To our knowledge, these are the highest efficiencies for as-cast solution-based devices employing non-fullerene acceptors.
