Family studies suggested that schizophrenia and bipolar disorders share some susceptibility factors. Schizotypal personality disorder may be an intermediate phenotype common to these disorders. Objectives : to study the specificity of schizotypal dimensions in relatives of schizophrenic or bipolar probands, to search an intrafamilial resemblance for these dimensions, and if an increased risk of schizophrenia and/or bipolar disorders is associated with a dimension in particular. Method : the study of the 3 schizotypal dimensions in a sample of first-degree relatives of schizophrenic and bipolar probands using the SPQ. Differences between groups were tested using a two-tailed t-test or ANOVA for continuous variables, a chi-squared test for discrete variables, the intra-familial correlation using intraclass correlation method, the familial risk using linear mixed models. Results : disorganization appears to be common to schizophrenia and psychotic bipolar disorders, and increases the familial risk of these two disorders. The negative dimension appears to be familial. Conclusions : disorganization appears to be a marker of susceptibility to psychosis, suggesting that the same genes are involved in schizophrenia and psychotic bipolar disorders. The familial nature of the negative dimension suggests that it is inherited. This implies that the quantitative phenotype definition and the study of more elementary phenotype dimensions should be used instead of nosological entities when performing genetic linkage and association studies in schizophrenia and bipolar disorders
