Proteins play an important role in cells via their enzymatic activity and the irinteractions. Their functions are mainly based on the protein structure. In order to detect their presence and to characterize their structure, we used optical properties of nanostructures. The localized surface plasmon resonance (LSPR), as well as the surface enhanced Raman scattering (SERS), allowed us to detect various proteins. We also optimized nanostructures to build a sensitive, reproducible and specific biosensor based on SERS. Indeed, specific detection of one pathological biomarker, the Manganese Super Oxide Dismutase (MnSOD) protein, was investigated by using optically optimized and aptamer-functionalized nanostructures. Using this system, we were able to detect the MnSOD at physiological concentration in body fluids, such as serum and saliva. Finally, the structural study of the Spleen Tyrosine kinase (Syk) protein by SERS, allowed us to demonstrate that its structure varied with its phosphorylation levels. A complementary Western Blot analysis showed that the Syk kinase activity depended also on its phosphorylation state, meaning that the structure and the activity of Syk were linked. Altogether, these data contributed to a better understanding of the interface between physics and biology

It reviews institutional developments and puts forward a comparative framework (PRECIS) describing behavioural insight teams with six key features. The report reaches four main conclusions: 1. in terms of capacity-building, there is significant dynamism and growing appetite to apply BIs to policy-making; 2. links between policy-making and academy communities can be strengthened and analysing large datasets offers great potential; 3. systematic application of BIs throughout the policy cycle can advance evidence-based policy-making; 4. need of more research on the long-term impacts of policy interventions

La mutation BRAFV600E est retrouvée dans plus de 80% des cas de leucémie à tricholeucocytes (HCL). Récemment des mutations additionnelles à BRAFV600E ont été mises en évidence. Un panel de 21 gènes d'intérêts (appelé Trichopanel) a été sélectionné à partir de la revue de la littérature des données de Whole Exome Sequencing. Nous avons analysé 20 patients atteints d'HCL et 4 formes variantes (HCL-v) au diagnostic (dont 5 paires diagnostic/rechute). Le Trichopanel a été informatif dans 96% (23/24) des patients et a révélé des mutations dans BRAF (n=18), KLF2 (n=4), MAP2K1 (n=3), KDM6A (n=2), CDKN1B (n=2), ARID1A (n=2), CREBBP (n=2) NOTCH1 (n=1) et ARID1B (n=1). Parmi les patients porteurs de la mutation BRAFV600E d'autres mutations ont été trouvées dans 33% des cas (6/18). Les fréquences alléliques de ces mutations étaient proche de celle de BRAFV600E suggérant qu'elles sont d'apparition précoce. Les 4 patients HCL-v présentaient des mutations dans les gènes de la régulation épigénétique dont KDM6A (n=2). Ces mutations induiraient la perte du domaine C-terminal, hautement conservé et essentiel pour l'activité déméthylase de la protéine. L'inactivation de KDM6A pourrait sensibiliser les cellules tumorales aux agents déméthylants tel que les inhibiteurs d'EZH2. Cinq patients (2HCL et 3 HCL-v) ont été analysés parallèlement au diagnostic et à la rechute/évolution. L'analyse des prélèvements a la rechute a montré une évolution clonale de la maladie. The workshop "Visions for the World in 2035" was organised in the EU Policy Lab by the Foresight and Behavioural Insights Unit in collaboration with the European Development Days (EDD15). It hosted 15 Future Leaders between the ages of 21 and 26 who were selected by the EDD15 organisers from around the world to join the debate on development policy and international cooperation. The purpose of the workshop was 1) to introduce the Future Leaders to systemic and long-term strategic thinking, 2) to help them formulate a vision broadly related to their area of activity and thus boost their contribution to the upcoming EDD15 debates, and 3) to foster their positioning as agents of positive change in the coming decades. Through group vision building, plenary discussions and individual pitches, participants shared their experiences, explored megatrends that are likely to shape different possible futures and challenged present assumptions related to social change, youth and development. The aim of the report is to present our insights from the preparation and running of the workshop in order to revise different methods and techniques used and adapted in the context of the EU Policy Lab. This type of an event could be further adapted and replicated in other research and policy contexts

La leucémie à tricholeucocytes (HCL) et sa forme variante (HCL-v) sont des syndromes lymphoprolifératifs B caractérisés par des mutations de BRAF et de MAP2K1 responsables d'une activation anormale de la voie des MAP-kinases. Des mutations additionnelles à ces anomalies drivers ont été identifiées. Nous nous sommes intéressés au gène KDM6A qui code pour une lysine déméthylase exerçant un rôle de suppresseur de tumeur. Sa perte d'expression sensibiliserait les cellules tumorales aux inhibiteurs d'EZH2, une méthyltransférase. Afin d'étudier l'impact fonctionnel de l'inactivation de KDM6A dans la HCL, nous avons transfecté un système CRISPR-Cas9 ciblant l'exon 4 du gène dans la lignée cellulaire JVM-3. La perte d'expression de KDM6A dans cette lignée favorise l'activation de la voie des MAPK sans modifier les capacités prolifératives des cellules, ni leur sensibilité aux inhibiteurs de BRAF. Les cellules éditées présentent une meilleure résistance à la privation en nutriments concordante avec une augmentation de l'expression de la survivine. Il n'a pas été observé de sensibilité accrue des cellules modifiées aux thérapies ciblant l'épigénétique incluant les inhibiteurs d'EZH2. L'inactivation de KDM6A paraît ainsi concourir à l'activation de la voie oncogénique des MAPK dans la HCL mais modifie peu les capacités des tricholeucocytes et leur réponse aux chimiothérapies et thérapies ciblées
