Forensic efficiency parameters for the 15 STR loci in the population of the island of Cres (Croatia)( )

Forensic parameters based on 15 AmpFISTR Identifiler short tandem repeat (STR)loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, VWA, TPOX, D18S51, D5S818 and FGA) were evaluated in the sample of 122 unrelated, autochthonous, adult individuals from the Island of Cres (Croatia). PCR amplification was performed with the AmpFISTR Identifiler PCR Amplification Kit and the amplified products were separated and detected using the ABI 3130 DNA genetic analyzer. The agreement with Hardy Weinberg Equilibrium (HWE) was confirmed for all loci (p> 0.05). The combined power ofdiscrimination (PD) and the combined power of exclusion (PE) for the 15 tested STR loci were 0.99999999999999997988728679 and 0.999997397, respectively. According to the presented data, D18S51 proved to be the most informative marker followed by markers D2S1338 and D21S11. Interpopulation comparisons in allele frequencies with other East Adriatic Islands revealed significant differences for all analyzed population pairs ranging from 4 loci (Cres vs. Hvar) to 1 locus (Cres vs. Krk). Furthermore, allele frequencies comparisons of Cres and Croatian mainland revealed the lack of statistically significant differences at all studied loci. The results of the current study indicate that the examined fifteen STR loci are useful genetic markers for individual identification and paternity testing in Croatian population from the Island of Cres
Toward Male Individualization with Rapidly Mutating Y-Chromosomal Short Tandem Repeats by Kaye N Ballantyne( )

Relevant for various areas of human genetics, Y-chromosomal short tandem repeats (Y-STRs) are commonly used for testing close paternal relationships among individuals and populations, and for male lineage identification. However, even the widely used 17-loci Yfiler set cannot resolve individuals and populations completely. Here, 52 centers generated quality-controlled data of 13 rapidly mutating (RM) Y-STRs in 14,644 related and unrelated males from 111 worldwide populations. Strikingly, greater than99% of the 12,272 unrelated males were completely individualized. Haplotype diversity was extremely high (global: 0.9999985, regional: 0.99836-0.9999988). Haplotype sharing between populations was almost absent except for six (0.05%) of the 12,156 haplotypes. Haplotype sharing within populations was generally rare (0.8% nonunique haplotypes), significantly lower in urban (0.9%) than rural (2.1%) and highest in endogamous groups (14.3%). Analysis of molecular variance revealed 99.98% of variation within populations, 0.018% among populations within groups, and 0.002% among groups. Of the 2,372 newly and 156 previously typed male relative pairs, 29% were differentiated including 27% of the 2,378 father-son pairs. Relative to Yfiler, haplotype diversity was increased in 86% of the populations tested and overall male relative differentiation was raised by 23.5%. Our study demonstrates the value of RMY-STRs in identifying and separating unrelated and related males and provides a reference database

Many historical episodes marked Bosnia and Herzegovina as a significant ethniccrossroads, which makes it a very interesting site for various population studies. The first stages of these complex investigations were based on observations of numerous phenotype markers. The following phase, which was relatively brief, was dominated by the use of different cytogenetic markers. Finally, at the beginning of this century, the molecular-genetic diversity of the BiH population became the focus of modern research. Autosomal and Y-STR markers, together with mitochondrial haplogroup (Hg) diversity were initially used in the examination of isolated groups, as well as the whole popu- lationof modern Bosnia and Herzegovina. The most recent study describes the distribution of Y-chromosome haplogroups in the three main ethnic groups in Bosnia and Herzegovina, and suggests a preliminary hypothesis for the process of peopling this area
Comparative study of two dna extraction methods in different tissues and conditions of degradation( )

Abstract: The aim of tis study is to compare two methods of extraction of DNA from different tissues (spleen, liver, lungs, kidney, hearth, mussles, and pancreas) with Ph-CL and QIAAamp ® DMNA Mini kit and to confirm which of this two methods give better results. Degradation of the tissues was in 3 controled conditions in a period of 6 months: room temperature, in a refridgerator on +4 and outside temperature in a period 2.3-7.8.2010 year. In total 210 analyses were performed and 20-30 mg of the tissye was taken. Quantification was performed with Quantifiler kit on 7500 RealTime PCR ABI and analysed with 7500 System SDS version 1, 2, 3 software. Continuosly temperature was measure with digital TESTO termomethers on each 2 h. Comparation of the two extraction protocols from the tissues was performed with T-test. Comparation of different tissues who were degradated in 3 controled conditions and extracted in 5 periods was performed with AMOVA. This study show that biger yeld of DNA from the tissues can be isolated with PhCl extraction method, but also with QIAAamp ® DNA Mini kit eaven there is les amount of isolated DNA it can be used for further PCR reaction. PhCl method its slow, cancerogenous, more expenicve and can be problem in amplification proces because of big amount of DNA. Comparative results from different tissues show that there is no big diferences in the extracted yeld of DNA. We proved that in putrefied tissues non toxic and fast method can be used for DNA extraction. Our study show that there in no decerasing of isolated DNA with increasing of PMI and ambiental temperature, maybe because of the short period of 6 months that we analyse. Althout in forensic DNA analysis we should take in mind that quality and quantity of extracted DNA depends of ambiental conditions, PMI and type of the tissue. The fastes degradation hapends in tissues who have more proteolotyc enzymes
