Detection of Chlamydia psittaci in cage birds in Slovenia by real-time PCR( )

Avian chlamydiosis is a zoonotic disease of birds caused by the bacterium Chlamydia psittaci. The highest infection rates are found in psittacine birds (Psittacidae) which are the most common cage birds. C. psittaci causes infections of the conjunctiva, respiratory tract, and digestive tract, with or without clinical signs in birds. Infected birds can shed chlamydiae through respiratory tract excretions and in faeces. Transmission of C. psittaci primarily occurs by close contact of infected bird to the susceptible bird or human. To determine the prevalence of C. psittaci in cage birds in Slovenia, oropharyngeal and cloacal swabs from 125 cage birds were examined by Chlamydiaceae-specific real-time polymerase chain reaction (real-time PCR). Two lovebirds (2/12) and a budgerigar (1/44) were positive for Chlamydiaceae by real-time PCR and were also positive for C. psittaci by an ompA-based real-time PCR assay specific for C. psittaci. Multiple loci variable number of tandem repeats analysis (MLVA) identified C. psittaci of genotype A in the positive budgerigar and C. psittaci of genotype B in the two positive lovebirds. The infected birds had no significant clinical signs of avian chlamydiosis on clinical examination. Using real-time PCR, the study showed a low prevalence (2.4 %) of C. psittaci in Slovenia

Evidence of avian influenza virus and paramyxovirus subtype 2 in wild-living passerine birds in Slovenia( )

A total of 670 cloacal swabs were taken from 37 species of wild-living passerine birds in years 2004, 2005, and 2006. The cloacal swabs were pooled into pools of three swabs for analysis. Isolation of avian influenza virus (AIV) and avian paramyxoviruses (APMV) was done on chicken embryos. One-step reverse-transcription polymerase chain reaction (RT-PCR) was used to detect AIV RNA. AIV nucleic acid was detected by RT-PCR in a sample of one common starling (Sturnus vulgaris). All attempts of AIV isolation from wild-living passerine birdsć samples were negative. APMV 2 was isolated in one robin (Erithacus rubecula) sample. In light of the presented results and literature data, passerines appear to play a minor role as potential disseminators of AIVand APMV; however, it seems that some wild-living passerine species could act as occasional carriers

S preiskavo vseh 23 priznanih vrst ptičjih mikoplazemsmo želeli ugotoviti ali imamo gena dnaK in groEL, ter določiti njuni zaporedji in izražanje. Z uporaboPCR za pomnožitev dnaK smo pri 21 vrstah ptičjih mikoplazem prvi določili zaporedja gena dnaK, ki so bila dolga 460-1400 nukleotidov. Analiza teh zaporedij s programom PHYLIP je pokazala enako filogenetsko razvrstitev v skupine, gruče in podgruče, kot jo za te mikoplazme kaže razvrstitev na osnovigenov 16S rRNA. S specifičnimi protitelesi mAB 3 smo sintezo proteina DnaK dokazali pri 31 vrstah mikoplazem, količina DnaK je bila primerljiva s količino EF-Tu. Zvišana temperatura pri vrsti Mycoplasma gallisepticum ali privrsti M. synoviae ni povečala sinteze DnaK, ki je za perutnino manj imunogen kot hemaglutinini pMGA in VIhA ter patogenih mikoplazem. Gen groEL jebilo mogoče dokazati le pri 5 vrstah skupine pneumoniae vključno z M. Gallisepticum in M. imitans. Za M. imitans smo prvi določili zaporedje groEL (neskončno850 nukleotidov), ki je bilo 93 % identično zaporedju M. gallisepticum. Ugotovili smo polimorfizme zaporedij dnaK in groEL M. gallisepticum in dnaK M. synoviae, vendar so bili manj številčnejši kot polimorfizmi njunih genov recA in vlhA
