EPR spin-probe oximetry of isolated skeletal muscle mitochondria by Lidija Honzak( )

The spin probe EPR -oximetry of isolated mitochondria from muscles of laboratory rats was used to study their functional states. We have found that EPR oximetry is sufficiently sensitive to follow the respiratory metabolic states 4-3 transitions in suspensions of mitochondria in spite of the low concentration of isolated mitochondria. Many advantages of this method compared with the classical respirometry for small samples with oxygen electrode, are evident. They suggest the use of EPR oximetry as a suitable method for studying the metabolic respiratory states of mitochondria in mitochondria dependent muscular myopathies. Small tissue samples provided by biopsies are suflicient to perform the measurements. The respiration of the sample containing mitochondria with different respiratory activity is discussed

The enhancement of topical delivery of hydrophilic substances by use of multilammelar liposomes was measured ex vivo on pig ear skin and in vivo on hairless mice by electron paramagnetic resonance method (EPR) Multilamellar liposomes with different lipid composition (final concentration of membrane components is 48 mg/ml) were loaded with a hydrophilic spin probe GluSL, whichdoes not penetrate the liposome membane easily. They were cheaclerized with respect to their slability, entrapped vulume and enhancement characteristics. We observed significant difierences in the pruperties of dilferent types of liposomw with rcspect to their stability when in conlact with the skin and fheir penetration intn the skin. 'fhe results measured in vivo arc consistent with those obtained cx vivo. On the basis of these findings the liposomes with appropriate stability and intradermal penetration characteristics were chosen for the developmem of liposome-encapsulated 1% clindamycin preparation for therapy of acne vulgaris. A double-blind clinical study was conducted to assess the safety and efficiency of liposome-encapsulated 1% clindamycin solution versus 1% clindamycin solution (Klimicin T, Lek). On the basis of the clinical trial or may be concluded thalliposome-encapsulared 1% clindamycin solution was therapeutically superiorever conventional 1% clindamycin solution in the treatment of acne vulgaris

In vivo low frequency EPR was used to measure the enhancement of topical delivery ot hydrophilic substances by use ot multilamellar liposomes. The contribution of transepidermal oržand transfollicular routes of transport was investigated using hairless and normal mice. Two liposome dispersions that previously had been shown to have different enhancement properties on ex vivo skin were used. The kinetics of the reduction of hydrophilic spin probe GluSL (N-(1-oxyl-2,2,6,6- tetramethyl-4-piperidinyl)-2,3,4,5,6-pentahydroxy-hexaneamide) applied to the skin encapsulated into the liposomes was measured. To distinguish the reduction of GluSL on the skin surface from its reduction inside the skinč theoxidizing agent potassium ferricyanide (KFeCN) was used. This does not penetrate into the skin and therefore it oxidizes hydroxylamines back to nitroxide only on the surface of the skin. We observed significant differencesin the properties of the two types of liposomes with respect to their stability when in contact with skin and their transport characteristics.The results measured in vivo are consistent with those obtained ex vivo, indicating that in vivo L-band EPR is a powerful technique for following pharmacokinetics in the skin of live animals. The results also show that clearance by blood flow and possible alterations of skin after sacrifice of animal do not influence the results of penetration of liposome entrapped substances into the skin during the time of our experiment (typically around 60 min). The reduction of GluSL in the skin of hairless vs. normal mice was similar, indicating that the transfollicular penetration was not of major importance in vivo in this experimental model

Mitochondria are the subcellular organelles where ATP is synthesized in the process of oxidative phosphorylation. As the high-energy electrons from NADH and FADH2 are transported down the respiratory chain in the mitochondrial inner membrane, the energy is released and is used to pump protons (H+) from the mitochondrial matrix to the intermembrane space. This creates an electrochemical proton gradient across the inner membrane of the mitochondria Back flow of H+ down this gradient is, in turn, used to drive the membrane-bound enzyme ATP synthase, which catalyses the formation of ATP. The respiration rate of isolated mitochondria is regulated by the ratio ATP/ADP. Pi. It was found that the respiration rate in vivo, is regulated intramuscularly in the same way (Brindle KM et al 1989). In different pathological situations, as well as extreme physiological trauma, hypoxia and even anoxia can occur. In this situation, ATP can be synthesized anaerobically. The end product of anaerobic metabolism is lactic acid, which lowers intramuscular pH. In some pathological situations, like ischemia, the diffusion of C02 which is produced in citric acid cycle in mitochondrial matrix is limited due to vasoconstriction. Higher concentrations of CO2, lowerpH of citosol and matrix. The aim of this work was to study the influenceof hypoxia and acidose on the oxidative phosphorylation of skeletal muscle mitochondria. Since the polarographic method with the Clark electrode is not sensitive enough for measurements of respiration rates in hypoxia, we developed the EPR-spin probe oximetry. We measured the respiration rate of isolated mitochondria and mitochondria in situ in muscle fibers. We measured the respiration rate of isolated mitochondria in buffers with different pH (7.4-6.2) and found that an acidic pH didn't influence the respiration rate.
